Diana,
If you know which lodge you think your relative was a member of you can find out if he was a Mason. Beginning in the 1840s, the Grand Lodge of Virginia published the proceedings of their annual meetings. On the last pages, there is a list of the members of each blue lodge for that year. The lodges are listed by name, but often included the name of the town where they were located.
What is your relative's name? I have quite a few of the lodge rosters from the antebellum period. If you are looking for someone from that era, I may have some information on him.
Many of the records are also available at public research libraries like UVA or the VHS, and as Lonny suggested at Grand Lodge of Virginia. Search for: "Proceedings of the Grand Annual Communication of the Grand Lodge of Virginia."
